You just put a piece of tail in about 100 ul 25 mM NaOH and 2 mM EDTA and boil 10 min. After boiling, vortex several seconds and add 100 ul 40 mM tris ph 7.4 and vortex again. Spin for 3-5 min and use for PCR. This is very quick; you will have PCR results in only a few hours of cutting the tail. Also, the proteinase K digest of tails is usually fine if done only several hours, overnight is not even necessary. We genotype all of our mice this way and it really is not harmful to the mice to have a tiny snippet of their tail taken. ------
